WebThe Tth DNA ligase is stable and active at much higher temperatures than conventional DNA ligases. The optimal ligation temperature range is 7-10 °C higher than that of the T4 DNA ligase and is determined by the Tm of the substrates. High ligation temperature eliminates the nonspecific ligation. WebIn order to minimize mismatch ligation, the minimal amount of ligase required for efficient ligation of matched substrates should be used. T4 and Tth ligase differ in cofactor requirement, but also in temperature and pH optima. T4 DNA ligase has a temperature optimum of 37°C, while Tth works best between 65-72°C (22,25–27).
